You can run two samples at the same time in different "wells" of the gel. It can never be reused to run the same well for a new sample. because the first sample may carries contaminating DNA or proteins that may affect the migration and authenticity of the next sample if run in the same well. And why you think to do this BTW. A gel has nowadays at least 10 wells. Run your 9 sample plus marker there, if you have more make a second gel and run!
