Iago opposes Othello because Othello chose to promote Cassio to lieutenant instead of Iago. Also, Iago is prejudice against Othello for his skin colour. Othello was a victim of the time he lived in.

Iago is convincing and lies to othello making him think that whatever iago is saying is true. He manages to con just about everybody around him.

In Othello's case, there is a tiny kernel of doubt imbedded in his humility. At the beginning of the play, this doubt is well-hidden, but it shows in Othello's "Most potent grave and reverend signiors" speech. He is truly amazed that someone as lovely and wonderful as Desdemona could love someone as old, battlescarred, and ugly as he is, someone with no title or position who isn't native to Venice. In that humble thought is the seed of the doubt that maybe she doesn't love him after all. Iago nurtures that seed until it bursts into full ugly flower.

It was always Iago's plan to destroy Othello. He gained Othello's trust with the specific intent to betray him later.

Why Iago hated Othello so much is unclear. He offers a number of reasons, none of them particularly persuasive.
