James Madison did not oppose the ratification of the constitution. In fact he encouraged it by helping to write the Federalist papers with Alexander Hamilton and John Jay.
Because some of them felt it would make the central government too strong and take away state rights. They were opposed to a strong central government, afraid it would be like Great Britain.
One notable figure who did not use a pseudonym to oppose the Constitution was George Mason. He was a prominent statesman and a delegate to the Constitutional Convention who ultimately refused to sign the Constitution due to his concerns over the lack of a Bill of Rights.
