Basically because several European countries - notably Portugal, the Dutch Republic and later England had opened the trade and transport of spices, silk and other precious commodities by sea. One sturdy trading vessel could carry as much as a whole large caravan with room to spare. And this new form of direct bulk trade and transport was moreover much cheaper and quicker than the overland long trek with hundreds of camels, camel drivers and all the middlemen involved in this route.

So basically, after the big trading ships arrived on the scene, the merchants of the silk route simply could no longer compete and their business dwindled.
