"Duh" is a smart-aleck-y way of saying something is obvious. For example, if you say "Wow, the sky is really blue" they might say "Duh!" instead of saying "Of course it's blue because that's the color the sky usually is." It's not really very polite, so you only say it in a very informal situation.
