Oliver and Orlando were sons of Sir Rowland De Boyz. However they both had completely different characters. While Orlando was gentle, caring and loving and had grown up to be a complete gentlemen, Oliver was cruel, tyrannical and merciless. As a result he didn't give Orlando any share of the money left by their father. However, below Oliver's cruel and mean nature, he knows that Orlando was a loving man and even thugh he had not had any education he had turned out a more 'educated' man than Oliver when he says,
"Yet he's gentle, never schooled and yet learned, full of noble device, of all sorts enchantingly beloved,and, indeed so much in the heart of the world, and especially of my own people, who best know him,that I am altogether misprized."
As a result, Oliver is very jealous of Orlando and that is why wants to harm him so that he is not over- shadowed by his brother.

Significance of opening scene in as you like it?
The opening scene of "As you like it" is basically a clumsy way of introducing the entire main cast to the audience. It introduces the characters of The dukes, Rosalind, Celia, Oliver and Orlando.
