Jacob is against Bella going on her honeymoon because he fears for her safety due to Edward being a vampire. He is also protective of Bella and doesn't want her to potentially get hurt or put in danger while on the honeymoon. Additionally, Jacob is still in love with Bella himself, so he may feel jealous or worried about her being with Edward in such an intimate setting.

Later on, they found out that Bella was going to have a baby that was half-human, half-vampire.
