I also have Windows Vista, and sometimes iTunes wont load either. It could be that something is installing or is waiting to be installed. That's what happened to me, something needed to be installed so on the start menu I clicked on a button and it has a lock symbol on it and it shut down and it installed. Then I started it back up and it still wouldn't work. I turned it off and logged in. Went into iTunes clicked on itunes store didn't work, so I tried it again AND IT WORKED!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!!

Instead of reading this whole thing you could: just log on and off a lot, and for the iTunes store if it says This could not complete the iTunes store request, just logg off and back on.
i am not sure why your apps are not loading but please try going to "Downloads" on your iTunes toolbar in iTunes and loading them there. Then sync your itouch with your itunes and they should transfer.

it probably has something to do with the internet, I don't have any problems with vista, try reopening it
