Scientists aren't really sure exactly what "causes" cancer, so it's difficult to answer this. Cancer is essentially a breakdown in cellular reproduction, causing the cells to reproduce out of control.
We know that exposure to certain chemicals ("carcinogens") increases the likelihood of cancer developing, but we also know that some people are exposed to those chemicals with no harmful effect and other people develop cancer for no clear reason.
That said, tobacco smoke (and smoke in general) does contain a lot of chemicals that are known carcinogens.
