Alexander Hamilton led the fight for ratification with support from John Jay over a 6-week period in Poughkeepsie, NY. It resulted in successful ratification on July 26, 1788 against strong opposition . The 11th state to do so.
James Madison did not oppose the ratification of the constitution. In fact he encouraged it by helping to write the Federalist papers with Alexander Hamilton and John Jay.

Alexander Hamilton, James Madison and John Jay (the three authors of The Federalist Papers) were important to the ratification in two key states: New York and Virginia. One major voice speaking out against ratification was Patrick Henry of Virginia (of "Give Me Liberty or Give Me Death" fame).

Together, the three wrote the Federalist papers, a collection of articles and writings pushing for a more Federalist government, and for the ratification of the Constitution. After the ratification, John Jay and Alexander Hamilton continued to support the Federalist party, Hamilton being the leader of it. Madison, however, joined with Jefferson in creating the Republican party and fiercely opposed the Hamilton plan. (this is 12 years after the ratification)
