Make sure the PSVita is fully updated and you've got the latest version of Content Manager Assistant installed on the PC, and that the program is currently running. Connect the PSVita to the PC by a USB cable and it should be detected, if not, might be a problem with the cable.
Or, you can enable wi-fi connection on Content Manager Assistant on the PC, and add the PC on the Vita's own Content Manager, that should allow connections over wi-fi.
