Why would you want to paint a turtle's shell anyway - unless it's for identification. A more sensible procedure would be micro-chipping ! It's permanent and doesn't wash off !
Anyway - to answer the question - so long as you use lead-free & solvent-free paint it shouldn't harm the turtle.

∙ 13y agono, if a turtles shell cracks it is most likely to grow moss over the crack to heel. However if the shell brakes then yes it will die. Because turtles are attached to their shell, and it is part of its body.
