This engine (3.0L) is an interference engine so yes, the engine could lock up if the timing belt jumped.
Loosen the bolt right below the tensioner screw and then tighten. Once it is tight enough, tighten the bolt back down and your done. Hello' Unless you are talking about the timing belt, there is not serpentine belt on a Lexus ES300. The timing belts is the one that runs thru the two camshaft, the water pump and the crankshaft. This belt can easily be tighten just like the above user say, however, be very carefull with the tensionomer, cause it has a pressed shaft on it that coul be dangerous.
There are pages and pages of things that can cause a check engine light. You need to have it checked with a scantool. Then the code can be diagnosed and repaired.

if your timing belt jumped one tooth or timing belt was installed incorrectly you will be out of sync also a defective timing belt tensioner will cause sync problems

The only reason the timing would have jumped is if the belt has lost some teeth which it will do right before it breaks. The timing belt should be replaced before the car is driven or the motor started again. If the belt breaks while the engine is running, it can cause major damage to the head and possibly the block.
Just had that problem on the rear of a Lexus ES300 and it turned out that both rear sway bar frame bushings were worn out. After replacement the car sounds quiet.
