Not sure, yet. Ubisoft has only mentioned that in Revelations, Desmond will be in a place called "The Black Room", where he is in an induced coma inside an Animus. Over the course of the game, he will try to piece back together his subconcious, and eventually regain conciousness.

However, she is able to see the future and knows that Desmond Miles is reliving Ezio's memories. With this knowledge, she explains that the world, and all of its inhabitants, were destroyed by a mysterious cataclysm. While fighting using "Right Trigger + X" to parry then attack (Xbox 360) will make you able to break their legs. There is no way to garuntee that will be the visual sequence used, but if you use the dagger (Short Blade) it increases your chances I believe. <--Assassins Creed 2

yes there's going to be one for sure. It comes out on November Ezio is still going to be the main character, I am not sure who else, but I know that Mario, other members of the Pazzi, Rodrigo lots of other assassins as well. You can train assassins and send them on to missions. You play as them during their missions, but if they die, they are gone forever. New weapons, such as guards will carry guns and Ezio will be able to do combo with his pistol as well. You can play as many different characters in mulitiplayer as well. Such as merchants, doctors, priests and courtseans. It's gonna be another fun and exciting game.
