Absolutely. Both North and South Korea have some of the largest militaries in terms of sheer size in the world, and South Korea is an economic power.
Now, if they properly united peacefully, without a devastating war wrecking the Korean economy, then we would then see the development of a new powerful regional power.
However, I think that this scenario is highly unlikely. Excluding outside interests, such as the Chinese and United States, that may not benefit from said unification, there is also the fact that the two Koreas are at heart so different.
Both have different languages and culture, not to mention that North Korea is behind South Korea in terms of infrastructure and economic development, which means that South Koreans will have to pay for future development in North Korea, as well as in their own respective territory. Not to mention international aid will cease for South Korea once the Koreas unite.
Overall, I think this scenario is highly unlikely largely due to the huge differences between the two leading parties in both countries.
However, it doesn't hurt to dream.

Probably if North Korea started a war, USA would go on and help South Korea for they are more friendly and because of similar government. China would help North Korea because of their similar government too. Japan would help South Korea because they are more of a close friend.
