No. It would make it awkward for you, your brother, and his friend, even if he does like you back. Believe me, I've been in this situation before, except my best friend liked my brother. And he liked her back. It just made it weird for everyone.
Because he's your 'friend' and not your 'boyfriend', so I guess it's okay for a guy 'friend' to to come up randomly and sit on your lap because friends are just random and weird like that. ;)

It is not weird but it is vaguely insulting to chose a friend simply because of their sexual orientation. It sounds almost like choosing a pair of shoes because they match your T-shirt. You chose friends primarily because you liked them, regardless of their sexual orientation.

In my expierience with relationships, it is always best to tell somebody straight away and then you can sort it out. If you leave it too long, it could develop and endanger your friendship.
