The plane with the largest wingspan in the world is the Stratolaunch. It has a wingspan of 385 feet.
Spruce Goose.
Howard Hughes, president of Hughes Aircraft Corporation, flew his plane, The H-4 Hercules nicknamed the Spruce Goose, in 1947. The Hercules (Spruce Goose) is the largest flying boat ever built, and has the largest wingspan of any aircraft in history. It survives in good condition at the Evergreem Avoatopm Museum in McMinnville, Oregon.
The spruce goose or H-4 Hercules is called the spruce goose because it was an aquatic plane and because it was made out of wood. The plane was actually made out of birch and not spruce but the press insisted on calling it that. Hope that answers your question ;)
Yes, the Spruce Goose is bigger and heavier than the B-52.
Wikipedia has a very nice page about the spruce goose. Additionally the Evergreen Aviation Museum in McMinnville OR, where the spruce goose is kept has a bit of info on their website. Both are linked below.

No, the Spruce Goose only ever flew once, it was a short test flight in November 1947.
Spruce is a tree name that rhymes with goose.
No, the Antonov An-225 is longer. The Spruce-Goose had a marginally wider wingspan and was the same height. But the A380 is the largest passenger aircraft.
