Your local auto parts store can sell you the tools needed to disconnect the heater hoses from the heater core. The tool comes in a set that is not expensive and is easy to use. You can also use the same tools for disconnecting fuel lines and AC connections.

Just gingerly pry it off.
The connector is of the Quick disconnect type. simpley Squeeze the the inner lock tabs with a pair of common pliers and twist while pulling.
On a 2001 Toyota Land Cruiser, the OBD-II connector is located under the dashboard on the driver's side. You can find it near the steering column, typically above the pedals. It may be necessary to remove a cover or panel to access the connector.
remove radio, heater control, glove box, dashboard trim & bezels. once you do this you can see a tublar bar which the dashboard secured to. remove the dashboard and the heater core is there in the center
If you have not done this before, it is a very time consuming job. Replacing the heater core is not the problem. Getting to it is. Unless you are willing to start removing panels and know how to remove them without damaging the plastic panels and clips, I suggest you take it to a professional. It is just a matter of removing enough to actually get to the core. Once you get to the heater core, first drain the radiator enough to get the level below the heater hoses. Remove the heater hoses, and then remove and replace the core.
If you mean heater core it's very easy...remove the right side cover under the radio 1 screw and a white bracket hold it in. Disconnect the two heater pipe clips and remove them, then pull out the heater core.
