If it was poured into the fuel tank it would sit on the bottom of the tank like sand. If some made it through the fuel pump pickup screen it would plug up the fuel filter.
You can't, you would have to drill a hole in the tank, spray water in it and let it drain. You would probably have to get a new fuel tank.
Assuming that there is a fuel filter in the fuel delivery system, it depends on how much water is in the tank, as well as where and how the fuel pickup tube is situated. Water will completely plug up a fuel filter and it takes surprisingly little water to do so (as little as a few mLs), but if the fuel pickup is not taking from the very bottom of the tank, a pretty significant amount of water would need to be in the tank to allow water to enter the fuel system.

If there is a significant amount of water in the fuel tank, then there will be a warning light.
The most effective way is to drain the fuel tank completely and then refill it with diesel fuel, add a bottle of methelated spirits to the fuel tank to ensure the water is no longer there, the metho breaks down the water particles and allows them to be burnt with the diesel, also change your fuel filter, as it can hold water, also the tank can become rusted /corroded due to the water being inside,
If you are getting water in the fuel filter, it is coming from your fuel tank. You will need to remove and clean the tank.
The fuel sending unit is located in the fuel tank itself. To get to it, you would have to drop your fuel tank, and you would remove it from the top of the fuel tank.
