No. Usually when you paint your wash your walls down first and rinse. Once the paint is applied it becomes a sealant. If it's water-based paint it's possible any pet odor (from a dog lifting their leg to a cat spraying on the wall) could be trapped in the plaster. After washing down the wall well (or that particular spot) use vinegar and warm water and soak lightly the part of the wall that has the stain and be sure it's good and dry before painting.
