No. There were several women who came before Earhart. Flight started in 1904 and Earhart was in the 1939 so there was 30+ years of flight before her. One was a woman named Ruth Wilson who flew in the National Air Races in 1928. Another was Harriet Quimby began flying in 1911 and was the first licensed woman who was licensed to fly. She also was the first woman to fly at night and over the English Channel. Quimby died in an airplane crash in 1912. Women flying dates back to the 1800's and in hot air balloons.

Earhart's first flying lesson was January 3, 1921 and received her license shortly afterwards. That summer she purchased her own second-hand Kinner Airster. She disappeared in 1937. Between 21 and 37, Earhart won many awards and competitions. She was an early female pilot, but not the first.
Baroness Raymonde de Laroche was the first woman to receive a first pilot's license in 1910 at Aero Club of France. Harriet Quimby was the first American, not the first in the world.
However, some credit Blanche Stuart Scott as the first American woman to fly, although she did so without permission and with no license. She basically took Glen Curtiss' airplane for a short hop without his permission. This occurred in Sept. 1910. So, some say Blanche was the first American, some say Quimby.
