For a bicycle:
1. Remove every component you do not want painted (recommend you remove everything but the frame)
2. If your bike already has a paint job you should take some 00 or 01 grit sandpaper to it to take off the clear coat and paint.
3. After your bike is sanded and ready to go prime it.
4. After priming (and letting it dry for at least 8 hours) your bike you can lay on the first coat of your desired color
5. Add at LEAST two more thin coats of your desired color
6. Hang to dry, or set in a dust free area to dry.
7. If you want a clear coat, apply in the same way after base color has dried
Technique:
Lay on spray paint coats in thin layers to avoid running (multiple thin layers is better than one or two thick layers)
Let each coat of paint dry COMPLETELY before you start the next one to avoid runs
DO NOT touch your piece while it is drying. Tie your hands if you have to. Touching a wet auto/bike paint job could result in nicking or scarring the paint job and would require starting all over again.
For a good paint job, you cannot cut corners. Be patient and you will be rewarded.
Personally, I wouldn't. Just about all paints that can be applied at home will be a lot softer than the factory paint, meaning that it will nick, chip and scratch a lot easier than the factory paint.
When I well and truly feel that it HAS TO be painted, I'll strip it, sand it down, mask it off halfheartedly and paint with hammerite and a brush. It's not particularly pretty, but at least the effort matches the result. Something that is rarely the case with a full DIY paintjob.

At home - with great difficulty. In a factory - reasonably easy. Tubing for frame is cut to length and joined. Frame is painted, then the bike is assembled.
