You have to remove the transmission pan. Although it is only a screen and not so much a filter it should not ever have to be replaced unless the transmission has failed completely.
there is a transmission dip stick, just like an oil dip stick. check with the dealer whether you should check the level when the car is off or running, and also if in neutral or drive?
Computer needs to see the button on the shifter so I would check the connections from this to the computer. If not then check fluid levels and then possibly have the fluid and filter changed. After that a mechanical linkage adjustment or internal issue.

I own a 2003 4 runner and the cabin filter is directly behind the glove box. Remove the glove box to get to it. Better yet, go buy the new cabin filter and check the box most come with instructions on how to change the filter
