The pedal clicks when it's down all the way, only hit that when you really need the acceleration. I burned a whole tank on it, and got a difference of about 2 MPG.
If you look on the dash of the Isuzu you should see a switch with an E on it. The E stands for economy. When this switch is on the transmission shifts differently, delivering better fuel economy.
Also with any car or truck. Keep the vehicle in good shape and service it regularly. A clogged air filter will cost you at the pump.
You can also gain 2 to 3 mpg by doing some research on the tires. Buy tires with less rolling resistance and keep the properly inflated.
Lastly if you are running a box style truck as opposed to a flatbed or utility body you can add an air dam on top of the cab to increase mpg. They are available on Ebay. I have a flatbed truck so I can't comment firsthand, but I see them in use on almost every semi truck on the road.

All above is true I would like to add that the air dams that can be purchased and installed on top of the cab do indeed increase mpg of the box style trucks. I know this first hand because talking to the owner/operator of a company which my work does all their service work was saying to me that even thou the addition is costly at approx $700-1500 depending on application it's worth it and pays for them selves within a few months of installation. He had estamated his fleet of 20+ NPR's were getting 5-8 more mpg with the air dam installed!! Seems low but add up the savings for an entire fleet and it's worth it and the only addition to the vehicle that actually showed a big savings and increase in mpg. He had tryed many other things with out the numbers adding up. So it might look funny, might make the cab heavier but is a great investment in increasing mpg. Do it!!
Another thing that wasn't noted above is to use your exhaust brake whenever possiable!! This will not only save on a little fuel but will also save your brakes too.
