There are actually three coolant drain plugs on the 2001 Tundra. Speaking of a 4.7 liter engine, there is one drain plug at the bottom of the driver's side of the radiator. The skid plate/splash shield need to be removed in order to access this. It is a plastic butterfly type and should be easily removed by hand or with the aid of pliers. The other two are located on the driver's side of the engine block.
