the answer in yes it would stay in one straight path as you release it. Until it loses more helium than the weight of the balloon. Then it will meander downwards til it flops of it's own weight.

Now, considering you are asking this question, I presume you can't undo knots otherwise you would have tried it yourself. If the balloon is filled with helium (the stuff that makes your voice higher pitched) then it should rise, but it would probably rise without the string being untied it that particular instance. If however you or someone else used their own breath to blow up the balloon then it will fall to the ground, which it will also do even if the string is attached (but only to the balloon mind).
