Something that's new, not cliché. Unique. Creative. And something people haven't thought of before. Dark vs. Light is always a good one, but can get boring after a while. Rather settle with something "grey" and brand new. Let's take the typical "zombie apocalypse" plot. It started with a virus that spread. What if it weren't the typical "random scientist makes a virus and accidentally lets it out," but rather some sort of evolved form of a common sickness? Small changes like that are alwaysinteresting.
Here's a good link to show you how writers come up with ideas -- being a good gamemaster is just like being a writer. You have to take ideas that are all around you and ask "what if?" to come up with a good roleplaying game scenario.
