Break off the red tab, then push the remaining piece all the way through. Save it, because you'll be putting it back, without that tab you broke off. You can now fill your transmission.
The dipstick is a shop dipstick, available online for $40-45. It does not stay in the tube permanently. You return the cap and push the red piece back in the direction you pushed it out.
