Dude if it's a 6 Cylinder then there are 6 plugs, 3 in the front and 3 in the back. The Ones In the back are the hardest to get, make sure you dont break one trying to get it out.
The 2000 Toyota Avalon spark plugs can be located on each side of the engine. The spark plugs will be near the bottom of the engine. There are three spark plugs on each side.
Flashing check engine lights means one or more cylinders are misfiring. Could be injectors, coils, plugs, wires, etc. Get it to a shop.

For both 4 & 6 cylinders, they are on the passenger side of the motor. Follow the plug wires to the plugs in the side of the engine.
It is probably your spark plugs misfiring.

The V6 would be ( 6 ) cylinders
There are 6 plugs 3 are on the driver side and 3 are on the passenger side alone with the coils
Same relative position as the front plugs. They are just a lot harder to reach.

Depends on the engine, the 2.0l and 2.4l options were 4 cylinders with 4 spark plugs, the 2.5 L Mitsubishi 6G73 V6 option required 6 spark plugs.
