alot of things maybey it got hit maybey it got wet maybey its old maybe you had an electric wire disconnect!
Try shooting a thin penetrant into the lock, and trying it. Do this daily for a week and if it doesn't work, replace the lock. not enough info. are you saying key doesn't work or manually locking by hand won't work, or both, key turns but door doesn't lock??? etc.

Depending on what year the car is the doors will automatically lock and what would cause them to unlock could be a faulty door latch There is a switch in the lock. probably that switch is broken in your car. If the inside lamp doesn't work when you open the door with that problem, that means the switch is broken.
To fix a door lock that won't turn, try lubricating the lock with graphite powder or WD-40. If that doesn't work, the lock may need to be replaced by a professional locksmith.
