"Best" is a matter of personal preference. Some people prefer interstates. Take I-264 W to I-95 south in Emporia to I-795 south to I-40 east in Piney Grove. That goes right into Wilmington. If I were driving, I'd take 13 south off of I-264 and take 11 south to I-40. It's about the same amount of driving time and it's 40 miles shorter. Personally, I think taking a yacht would be "best."
Take I-95 SOUTH. Make sure you bypass Providence by taking I-295 (EXIT 4 off I-95 in Massachusetts).
I would estimate about 5 hours (give or take 10 minutes). The best route would be to take A1(M) to A1 to A17 to A47.
To get to Wilmington, DE from Lancaster, PA, you can take Route 30 East towards Coatesville, then merge onto I-95 South. Follow I-95 for about 30 miles, and take the exit for Wilmington. The drive typically takes around 1 to 1.5 hours, depending on traffic. Alternatively, you can take a train from Lancaster to Wilmington for a convenient option.
