No. New tires does not change alignment settings as alignment settings done with tie rods, camber and caster bolts and not affecetd after tyre changing. Yaa But it should taken under consideration that after changing tyres its better to check alignment because there can be little change in alignment readings due to difference in tyres depths of old and new tyres.

New tyres change balancing settings so wheel balincing is must check after changing tyres.

A four wheel alignment can be done with new ones on the front and old ones on the back. Rotating them at some point will not affect the alignment.
