If one of the colonies was to be contaminated by something in air, you would be able to tell by there being a difference in look for specimen. Also looking to see if something is growing on top of smooth specimen.

The best way to sub culturing is by picking the colony with loop and streak it on fresh plate by streak plate method in different patterns another way to check the contaminatoion is swapin the colony, in this first take dd water which should be autoclaved properly and then dissolve a part of colony in it. Now with the help of sawaber or by pouring the solution on fresh plate inoculate the colonial solution.
If u find morphologically different colony then it is contaminated
The quick method is to do a staining but it is not relaible every time as many cells have same staining properties so perform both of them for best result.
