Personally, I prefer to gram stain the bacteria. However other like to find out what it's respiratory orientation is first (whether it is anaerobic, facilitative or aerobic).
There is really no standard way, just do what you feel is best, as long as you do all of them at some point.
The first step in inserting a new gene into a bacterium is to isolate the gene of interest and prepare it for insertion. This can involve cutting the gene with restriction enzymes and ligating it into a vector, which is a piece of DNA that can deliver the gene into the bacterium.
Politely but firmly ask the individual to step back and inform them that they need proper identification to access the area. If the individual becomes irate, remain calm and contact security or a supervisor to handle the situation. Do not engage in a confrontation.
Nomination may not be the first step in a process when there are prerequisites that need to be completed before the nomination can take place. This could include meeting specific criteria, obtaining approvals, or completing certain tasks before being nominated. In these cases, the critical first step would be completing these prerequisites before moving on to the nomination stage.
The first step to respiration is glycolysis.
The first step in chemical digestion occurs in the mouth with the enzyme amylase breaking down carbohydrates into simpler sugars.

Problem and opportunity identification
the first step is always the hypothesis but i say it's the identification of the problem which comes first... that's what they taught us in school...

The first step in the policy cycle is usually problem identification and agenda setting. This involves recognizing issues that require attention, defining them as problems, and placing them on the agenda for policy development.
Step one is by expressing one of the equation into one term that is taking one unknown in the form of other. Step two is replacing the unknown into equation 2. Step 3 is replacing the found unknown into one of initial equations to find the other unknown.
