Negative staining techniques are designed to stain everything BUT the bacterial cells. This allows us to see the cells unstained and helps us observe their morphology (how the cells are shaped and how they group together). The cells have an overall negative charge on their surface, so they naturally attract positive charges. Crystal Violet carries a positive charge, so it would adhere to the surfaces of the cells thus staining the cells (which is not what you want in a negative stain!)
Gram-staining does not stain the endospore due to the tough, resistant water-proof structure. It appears as an unstained area in a vegetative cell. Malachite green must be forced into the endospore with heat to stain it.
Specimen
A CBC specimen may be rejected due to specimen hemolysis (breakdown of red blood cells), inadequate sample volume, clotted specimen, improper handling or storage, or outdated sample. These factors can affect the accuracy and reliability of the test results.
There are two types of stains, the simple stain and the differential stain. A simple stain colors all objects the same while a differential stain is used to spot differences in microorganisms. A gram stain is a differential stain, which is used to tell the difference in gram negative and gram positive bacteria. A simple stain would stain all the organisms the same and this difference would not be noted. You would be able to determine their shape, whether it is a cocci or bacillus (rod), but not the type. I'm not sure why the simple stain would be preferable unless you just wanted a quick answer as to the shape of the bacteria. In some cases, a wet prep can be made of a presumptive gram positive cocci to tell the difference between bacteria or yeast. Otherwise, I would say that the gram stain is the only way to go.

If a Gram stain had been performed instead of an endospore stain, the slide would show the bacterial cells either as purple (Gram-positive) or pink (Gram-negative) based on their cell wall composition. The endospores, which typically do not retain the Gram stain, would likely appear as clear or colorless structures within the stained cells. Thus, the focus would be on the overall morphology and arrangement of the vegetative cells rather than highlighting the presence of endospores.

Specimen
Collecting a urine specimen from a urine drainage bag is acceptable when it is necessary to obtain a sample for laboratory analysis, and no other options are available, such as a clean catch. However, it should be done with caution to avoid contamination, and the specimen should be collected from the sampling port rather than the bag itself. It is important to ensure the sample is fresh and representative of the patient’s urine output. In general practice, obtaining a sample directly from the patient is preferred when possible.

If no heat fixing was done to a slide with a specimen on it, it would be rinsed off with the gram staining procedure. Heat fixing the specimen does kill specimen but it also locks it in place.
