By constructing a calibration curve (e.g. Absorbance vs a series of known standards) you overcome the problem of non-linearity within the measurement range. If you only take a single calibration point ( plus a zero point) you assume that the calibration curve is precisely linear with zero point intercept, which is not always the case.
