It really depends on the sound system you're using. Some are designed to handle loud volumes without destroying the speakers and others are not. In general, a speaker does wear as it's being used and yes, it is possible to ruin a set of speakers by playing them too loudly.
How will you know? well, one of the first signs is that the music starts sounding fuzzy, or has a distorted sound all the time when they're being played at a significant volume.
Of course, don't discount the quality of the music you're playing, as that has a significant factor, too.
