If you're just trying to play sounds through your mics input, you need to enable wave out/stereo mix on your soundcard.

If you can't find either wave out or stereo mix then your soundcard might not support them, sorry.

Once that's enabled all audio you play will stream through your microphone.

Can computer program in c plus plus be made speak letters?

Yes. Record WAV files for each letter, trim them to remove silent leads and trailers, then compress them using MP3 or similar codec. Your program then responds to input by playing the appropriate file. Obviously this is somewhat simplistic, but once you have the basic program in place you can embellish it further with more complex sound files to produce a more human-like vocabulary, by combining sound files to make words.
