This is a slightly controversial subject in southern England! The Mayflower departed from Southampton but then returned to Plymouth while a sister ship was repaired. The Mayflower then departed from Plymouth around a month later. So you can argue for either Southampton or Plymouth.
To add to the confusion, the Mayflower then stopped off at Newlyn in Cornwall for water.
