The best way for a family of 4 to get to Anna Maria Island would depend on where you were coming from - since I suspect that you will be coming from some other part of the country you would do best to fly into Tampa and then rent a car. From the airport take I-275 through St. Petersburg to US 41 south to Manatee Avenue west to where it ends on Gulf Dr. where a right turn will take you to Holmes Beach which is where Anna Maria Island is located.
