A mesocyclone forms withing a thunderstorm as a result of wind shear, or a change in wind speed and direction with altitude. For a northern Hemisphere supercell this would normally involve wind near the ground coming out of the west while stronger winds at higher altitudes come out of the south, creating a counterclockwise shift in wind direction with increasing altitude. The causes the updraft of a thunderstorm (in which air rises) to take on a counterclockwise rotation. The process is similar in the southern hemisphere, only there the upper level winds are out of the north, resulting in clockwise rotation.

If the shear is strong enough it can essentially tilt a thunderstorm, this separates the updraft and downdraft of the thunderstorm, preventing them from interfering with one another.
