There are two words for "to understand" in Spanish. These are entender and comprender. You could ask the question in two ways, depending on whether you are speaking to a friend-family member or a unfamiliar person, or a superior. The familiar would be ¿Entiendes? (¿Comprendes?). The other, formal form would be ¿Entiende usted? (¿Comprende usted?). The usted is not totally necessary, but makes it more formal.
