Generally not. If it is easily rotated, it probably means that the fittings are not quite secure. However, it should be noted that many drains, especially the older iron ones are not air-tight. As long as you get a relatively good seal and water doesn't come gushing out when you get a clog, you are probably okay.

Do the wheels of a car rotate backwards?

Unless a brake is engaged, the wheels on a car in neutral can rotate backwards. If the car is in gear, wheels attached to the power train probably will only rotate backwards if the car is in reverse gear. If it is in forward gear or "park" they should not rotate backwards. Wheels not connected to the power train should be able to rotate either direction.

Should front tires spin when the vehicle is jacked up high enough that the tires no longer touch the ground?

If the vehicle is a rear-wheel drive, the tires should be able to rotate when not in contact with the ground - unless the brake is set to stop the rotation. If it is a front-wheel drive, the tires should not rotate unless the vehicle is in neutral - likewise if 4-wheel drive is engaged.
