The "S" stands for Sonderklasse, which essentially means "special", in the sense of superior, or super. The "E" stands for Einspritz, that is, fuel injection. You could have one without the other, for instance I had a 1958 220S, a gussied up version of the 220. A 220SE was also available, top of the line.
