If you mean male and female, then men have a penis, testes, and other organs which are all designed to impregnate a female. Women have a vagina, ovaries, a uterus, a cervix, and fallopian tubes, and that is for being able to become pregnant. Women also have mature breasts, while nearly everyone has breast tissue (it is not usually developed in men unless they have gynecomastia, a condition where men grow female breasts). Men generally have larger muscles, larger size, sharper facial bone features, and a deeper voice when compared to woman. Men also tend to have narrower pelvises and the opening in the floor is more triangular rather than oval like in woman.

If you mean man and woman to refer to who they are (gender identity) and which gender culture they have (gender role, gender expression), then you can tell that by attitude, dress, and way of looking at life. Those who identify as men dress and act as men. In the US, if someone is wearing a dress, earrings, feminine hair, makeup, jewelry, etc., it means they think of themselves as female and expect others to treat them as such. Knowing what body parts they have is irrelevant. Similar goes if someone is growing a beard, appears to have no breasts, has short hair, and wears jeans and shirts from the men's section, and doesn't wear jewelry. They would not do that if they didn't want to be treated as men. Of course, some men and women do choose a few items from the other gender category, such as men who wear earrings and women who have short hair. Single differences like that from prevailing gender culture doesn't mean someone wants to be treated as the other gender.
