Defensive maneuvers. Generally it would be behind the knee-caps or really any major ligament of the body that is part of a major limbs (i.e. the elbow, the ankle, the wrist, etc.) In other cases such as in males the testicles would be the most sensitive depending on the strength of the man but if your trying to get him on the ground the knee-caps or behind the knee-caps would be a better choice.
Physiology. For touch sensitivity, as in the least amount of force to elicit a verbal response, such as "I felt that", the face, especially the lips, nose and eyes would be the major contenders. Sensitivity to touch level stimuli is not the same as sensitivity to pain. For example, with sunburn touch sensitivity decreases whereas pain sensitivity increases! If you mean inside the vagina, it is called the G-spot, and not every woman has that. If you mean on the top of the vulva, the sensitive spot there is called the clitoris. It is sort of like a tiny penis, and it gets erect during arousal. It is possible that the G-spot mentioned above is the part of the vagina wall that pushes against the nerves leading to the clitoris, thus the clitoris gets stimulated from the inside while it is engorged.
