As Macduff was going to wake King Duncan he discovered that King Duncan had been killed.
Macduff and Lennox appeared at Inverness Castle, which was Macbeth's home. Macduff indicated that the King had wanted him to stop by early. Macbeth led Macduff to the room where the King was staying. But only Macduff went inside. Macduff therefore was first to discover the murdered King Duncan.
Macduff is the first person to discover that the king has been murdered. He is therefore the first person to react to the death.
Malcolm told Macduff that he was greedy, and lustful and had absolutely no desire to be king.
