When Romeo is banished from Verona he goes to Mantua.

Friar Lawrence advises Romeo to go to Mantua until things have settled down in Verona. He believes that staying away for a while will help defuse the tension and allow for a better outcome for Romeo and Juliet.
In fair Verona, where we lay our scene....this is part of the prologue at the very beginning of Romeo and Juliet and it says the setting is in Verona, Italy where the story takes place. When Romeo gets exiled he goes to a place called Mantua.
Balthasar goes to Mantua to deliver Romeo's message about Juliet's faked death, in order to prevent Romeo from committing suicide when he hears the news of her death.

At the beginning of the act, he is in Juliet's bedchamber before he goes into exile in Mantua for the rest of the act.
