Because the grains are different, they can make the wood difficult to work with. The knothole may even fall out of the lumber as it dries, leaving a natural hole in the wood. If someone where to call you a knothole, they are saying your are flawed, like the wood.
It is not a species of tree- it is a tree that has a small hollow where a branch died. Where branches grow, it leaves a round marking in the grain of the wood- a "knot". If the knot falls out, you have a knothole.
