I've read that you can begin distinguishing the fish at about one week of age. this is very hard (for me at least because they like to squirm around) and also somewhat unnecessary. waiting until they are about a month old is easier and also around the time you can begin separating them if you chose to do so. a Female will have a larger more fan-like fin on it's underside and a male will have a modified fin called a "gonopodium".
The Gravid female will have the dark "gravid spot" in front of her vent. I can however assure you that if there has been a male in the same tank with a female guppy for any reasonable length of time she will be gravid.

You can tell by it's anal fin that is located at the bottom of the fish. If it is a male guppy it will have a gonopodium it looks like a backwards point, and if it is a female it will have a anal fin that looks like a backwards Chinese or Japanese fan.
